Development of diagnostic and prophylactic methodologies is dependent on knowledge of the host’s defence system and reaction to different vaccine adjuvants. Here we present a sequential morphological study of peritonitis and inflammatory cell processing of incomplete Freund’s adjuvant (IFA) in intraperitoneally injected Atlantic cod. The peritoneal tissue responses were characterised using necropsy, histology and electron microscopy. An extensive inflammatory response as characterised by leukocyte morphology and contents of enzymes, presence of apoptotic cells and IFN-γ-expressing cells was observed. Three days post injection, IFA droplets were surrounded by different types of inflammatory cells and two different patterns could be discerned. The first was characterised by flattened and concentrically arranged interdigitating cells connected by desmosomes and with macrophage-like cells (MLCs) predominant in the periphery. The second type possessed four stratified layers with an inner layer containing many apoptotic MLCs; a second layer containing flattened and shrunken cells and outer layers comprising moderately flattened cells and an outermost layer of mononuclear cells expressing IFN-γ. Oil was detected both inside and outside MLCs. The two types of processes, of which the second was clearly stratified, were similar to those observed in other teleosts, indicating a variety of reaction modes or alternatively sequential process development. The numerous dead MLCs contributed to inflammation.  相似文献

The western flower thrips, Frankliniella occidentalis (Pergande) (Thysanoptera: Thripidae), causes major losses in agricultural and horticultural crops worldwide. The volatile compounds methyl isonicotinate, p‐anisaldehyde, eugenol, and linalool are known as olfactory attractants, and salicylaldehyde is known as a repellent for F. occidentalis under clean‐air conditions in laboratory experiments. In the present study we assessed the responses of F. occidentalis to these compounds when presented alone, in combination, and in the presence of background odours emanating from cucumber (Cucumis sativus L., Cucurbitaceae), capsicum (Capiscum anuum L., Solanaceae), chrysanthemum (Chrysanthemum morifolium Ramat., Asteraceae), clove basil (Ocimum gratissimum L.), and lavender (Lavandula angustifolia L., both Lamiaceae) plants in a Y‐tube olfactometer. In the presence of any background plant odour, the attractiveness of pure methyl isonicotinate to F. occidentalis proved to be consistently significant. Compared to clean‐air conditions, a slightly lower percentage of thrips chose the Y‐tube arm loaded with 10% p‐anisaldehyde in the presence of cucumber leaf odour. With non‐flowering clove basil plants in the background, F. occidentalis responses to 1% eugenol, a constituent of clove basil essential oil, were neutral, and the same applied to responses to pure linalool, a constituent of lavender essential oil, in the presence of flowering lavender plants. Also, thrips responses to pure or diluted salicylaldehyde were clearly influenced by plant background odours. We simulated a push‐pull situation and found a trend indicating that the percentage of F. occidentalis choosing the airflow loaded with the attractant methyl isonicotinate was higher when the airflow in the other arm of the Y‐tube was loaded with the repellent salicylaldehyde compared to clean air, and vice versa. We showed interactions between attractive or repellent volatile compounds and the environmental odours in the chemical ecology of F. occidentalis and the potential of a combined use of these compounds in thrips pest management.  相似文献

Many Arctic regions are currently experiencing substantial summer and winter climate changes. Litter decomposition is a fundamental component of ecosystem carbon and nutrient cycles, with fungi being among the primary decomposers. To assess the impacts of seasonal climatic changes on litter fungal communities and their functioning, Betula glandulosa leaf litter was surface‐incubated in two adjacent low Arctic sites with contrasting soil moisture regimes: dry shrub heath and wet sedge tundra at Disko Island, Greenland. At both sites, we investigated the impacts of factorial combinations of enhanced summer warming (using open‐top chambers; OTCs) and deepened snow (using snow fences) on surface litter mass loss, chemistry and fungal decomposer communities after approximately 1 year. Enhanced summer warming significantly restricted litter mass loss by 32% in the dry and 17% in the wet site. Litter moisture content was significantly reduced by summer warming in the dry, but not in the wet site. Likewise, fungal total abundance and diversity were reduced by OTC warming at the dry site, while comparatively modest warming effects were observed in the wet site. These results suggest that increased evapotranspiration in the OTC plots lowered litter moisture content to the point where fungal decomposition activities became inhibited. In contrast, snow addition enhanced fungal abundance in both sites but did not significantly affect litter mass loss rates. Across sites, control plots only shared 15% of their fungal phylotypes, suggesting strong local controls on fungal decomposer community composition. Nevertheless, fungal community functioning (litter decomposition) was negatively affected by warming in both sites. We conclude that although buried soil organic matter decomposition is widely expected to increase with future summer warming, surface litter decay and nutrient turnover rates in both xeric and relatively moist tundra are likely to be significantly restricted by the evaporative drying associated with warmer air temperatures.  相似文献

Apoptosis is increasingly implicated as an early line of defense against viral infections. Viruses have devised numerous strategies to delay apoptosis of infected cells. Many viruses encode cell death suppressors that target mitochondrial apoptotic signaling pathway, indicating the importance of this pathway in the anti-viral response. Human and primate cytomegaloviruses encode the viral mitochondria-localized inhibitor of apoptosis vMIA, but no overt homologue of vMIA was identified in any non-primate cytomegalovirus. Here we report that m38.5 protein encoded by murine cytomegalovirus, which is unrelated to vMIA in its amino acid sequence, delays death receptor ligation-induced cell death, and that m38.5 associates with Bax, recruits it to mitochondria, and blocks Bax-mediated but not Bak-mediated mitochondrial outer membrane permeabilization. Thus, primate and murine cytomegaloviruses have evolved non-homologous but functionally similar cell death suppressors selectively targeting the Bax-mediated branch of the mitochondrial apoptotic signaling pathway, indicating the importance of this branch in the response of diverse host organisms against cytomegalovirus infections.  相似文献

During the course of screening Bacillus species from food and water in Norway, we isolated a strain of Bacillus sphaericus of DNA homology group V, not previously recognized to contain entomopathogenic strains, that was cytotoxic to Vero cell epithelia. Peptide mass fingerprinting of a protein purified from the culture supernatant of B. sphaericus B354 identified a cholesterol-dependent cytolysin (CDC) with high amino acid sequence identity with sphaericolysin, a CDC identified recently in B. sphaericus DNA homology group IIA. The toxin was haemolytic against erythrocytes from a range of species. Haemolysis was potentiated by dithiothreitol and inhibited by preincubation with cholesterol. The toxin induced lactate dehydrogenase release from Vero cells and formed pores in planar lipid bilayers. The distribution of CDC genes in B. sphaericus was examined, with CDC gene products obtained in 13 out of 17 strains representing four of the six DNA homology groups. Thus, we demonstrate the presence of a CDC in a nonentomopathogenic DNA homology group of B. sphaericus (group V) with typical CDC characteristics. CDCs appear to be present in a high proportion of B. sphaericus strains and are not restricted to group IIA insecticidal strains.  相似文献

The Manila clam Ruditapes philippinarum was introduced to Norway in 1987 and was produced in 2 hatcheries until 1991. Clam seed was planted at 6 sites. Two sites were on the Island of Tysnes, south of Bergen. Surviving adult Manila clams were recovered in 1995 and 1996. In the present study, Manila clams from the original seeding that displayed morphological signs of brown ring disease (BRD) were recovered in June 2003 (n=7) and in June 2004 (n=17). Samples from extrapallial fluid, tissues and haemolymph were inoculated on marine agar. Replicate subcultures on selective media were used to select potential Vibrio tapetis strains, and in total, 190 bacterial strains were isolated. One of these strains clustered within the V tapetis clade and was named NRP 45. DNA:DNA hybridisation with the type strain CECT4600 showed 52.7 and 57.3% DNA:DNA similarity. Hybridisation of NRP 45 and the V tapetis LP2 strain, isolated from corkwing wrasse Symphodus melops, produced 46.6 and 44.4% re-association. Partial gene segments encoding 16S rRNA, gyrase B protein (GyrB) and chaperonin 60 protein (Cpn60) were characterised and compared to CECT 4600. NRP 45 showed 5 differences in the 1416 nucleotides (nt) of the 16S rRNA encoding gene (99.6% similarity), while the GyrB encoding gene had 62 substitutions of 1181 nt compared (94.8% similarity) and the Cpn60 encoding gene had 22 substitutions out of 548 nt compared (96% similarity). This is the first finding of BRD and the first isolation of a V. tapetis-like bacterial strain from a bivalve in Norway.  相似文献

Fusobacterium nucleatum is associated with periodontitis in humans, and is a central member of the dental biofilm. Heat shock proteins (HSPs) of many different bacteria have been considered to play important roles during inflammations and infections. We have identified and characterised the HSP60 and HSP70, the Escherichia coli GroEL and DnaK homologues, respectively, in F. nucleatum ATCC 10953. The N-terminal 22 amino acid residues of HSP60 exhibited up to 63.6% identity with members of the HSP60 heat shock protein family of some selected bacterial species, while the N-terminal of 25 residues of HSP70 revealed up to 80% identity with members of the HSP70 family. The subcellular localisation of HSP60 and HSP70 was analysed by immunoblotting of bacterial cell fractions and immunoelectron microscopy of whole cells. HSP60 and HSP70 were localised in the cytosol, associated with membranes and extracellular fractions. These results are consistent with localisation for HSPs found in other micro-organisms, which further lead to the suggestion of a potential role in the pathogenesis of infectious diseases.  相似文献
